چکیده

This paper presents a diffusion experimental study for ternary lysozymeNa2SO4-water system, from moderate precipitant concentrations into the supersaturated region and provides a complete set of four diffusion coefficients. These data are important in order to provide accurate models of protein diffusion with applications in growth of protein crystals for X-ray diffraction studies. All three-component mutualdiffusion experiments reported here were performed by Rayleigh interferometry at pH= 4.5, T= 25 C and at a mean lysozyme concentration (average of top and bottom solution concentrations) of 0.6 mM (8.6 mg/mL). Four experiments, with different combinations of protein and Na2SO4 concentration differences, were performed at each of five mean Na2SO4 concentrations (0.1, 0.25, 0.5, 0.65 and 0.8 M), for a total of 20 experiments. In addition, we have measured dynamic light-scattering diffusion coefficients of the ternary system lysozyme chloride-Na2SO4-water. Precision Measurements of Binary and Multicomponent Diffusion Coefficients in Protein Solutions Relevant to Crystal Growth: Lysozyme Chloride in Water and Aqueous NaCl at pH 4.5 and 25 °C⊥

Accurate models of protein diffusion are important in a number of applications, including liquidliquid phase separation and growth of protein crystals for X-ray diffraction studies. In concentrated multicomponent protein systems, significant deviations from pseudobinary behavior can be expected. Rayleigh interferometry is used to measure the four elements (Dij)v of the ternary diffusion coeffic...

Extraction of Thermodynamic Data from Ternary Diffusion Coefficients of Lysozyme Chloride in Water and Aqueous Na2SO4

This paper presents, for ternary lysozyme-Na2SO4-water system, the thermodynamic data extracted from the measured values of four ternary diffusion coefficients and the Onsager reciprocal relations. The calculation for derivatives of solute chemical potentials with respect to solute molar concentrations was made using the method presented in [1]. This method is applicable to systems in which the...

Multicomponent diffusion of lysozyme in aqueous calcium chloride. The role of common-ion effects and protein-salt preferential interactions.

To investigate the effect of calcium salts on the thermodynamic and transport properties of aqueous solutions of proteins, we report ternary diffusion coefficients for the lysozyme-CaCl2-water ternary system at 25 degrees C and pH 4.5. We have used our ternary diffusion coefficients to calculate preferential-interaction coefficients as a function of salt concentration. This has allowed us to ch...

The effect of salt stoichiometry on protein-salt interactions determined by ternary diffusion in aqueous solutions.

We report the four diffusion coefficients for the lysozyme-MgCl2-water ternary system at 25 degrees C and pH 4.5. The comparison with previous results for the lysozyme-NaCl-water ternary system is used to examine the effect of salt stoichiometry on the transport properties of lysozyme-salt aqueous mixtures. We find that the two cross-diffusion coefficients are very sensitive to salt stoichiomet...

The effect of salt on protein chemical potential determined by ternary diffusion in aqueous solutions.

We use accurate thermodynamic derivatives extracted from high-precision measurements of the four volume-fixed diffusion coefficients in ternary solutions of lysozyme chloride in aqueous NaCl, NH4Cl, and KCl at pH 4.5 and 25 degrees C to (a) assess the relative contributions of the common-ion and nonideality effects to the protein chemical potential as a function of salt concentration, (b) compa...
